Swine farm wastewater discharge causes masculinization of western mosquitofish (Gambusia affinis).
Abstract Natural and synthetic steroid hormones have been detected in swine farm wastewaters (SFWs) and receiving waters. However, little is known on their potential endocrine disrupting effects on fish population in receiving rivers. Here we investigated the concentrations of androgens and estrogens in the SFWs and the receiving waters. In addition, the endocrine disrupting effects of SFWs on wild western mosquitofish (Gambusia affinis) were also analyzed by assessing the sex ratio, secondary sex characteristics and transcriptional expression of genes related to the endocrine system. Chemical analysis showed the ...

Endocrine disruption in western mosquitofish from open and closed aquatic ecosystems polluted by swine farm wastewaters.
In this study, a total number of 3078 adult western mosquitofish (Gambusia affinis) were collected and the sex ratio was determined. In addition, secondary sexual characteristics were examined and the transcriptional levels of target genes were analyzed. The results showed the mosquitofish populations had a significant increase in male-to-female ratio from 7 sites (including 2 fish ponds) among the 9 sampling sites. The hemal spines of females were masculinized at most sites while the hemal spines of males were feminized at approximately half of the sites (including 2 fish ponds). Significant reduction in vitellogenin (Vtg...

In search for globally disordered apo-parvalbumins: Case of parvalbumin β-1 from coho salmon.
Abstract Parvalbumin (PA) is a classical EF-hand calcium-binding protein of muscle, neuronal, and other tissues, and a major fish allergen. Although certain apo-PAs lack tertiary structure, functional implications of that feature and its structural prerequisites remain unclear. In a search for unstable PAs, we probed conformational stability of parvalbumin β-1 from coho salmon (csPA), a cold water fish species, using circular dichroism, scanning calorimetry, hydrophobic probe fluorescence, limited proteolysis, chemical crosslinking and dynamic light scattering techniques. Apo-csPA is shown to be mainly monomeric ...

Don ’t Be Afraid to Search in the Dark: Jon Lorsch Encourages Graduates to Consider New Perspectives
Jon Lorsch, from Swarthmore College’s class of 1990, returned to his alma mater in May to accept an honorary Doctor of Sciences degree for his accomplishments as a biochemist and his visionary leadership of NIGMS. During the university’s 147th commencement, he spoke to the 2019 graduating class, offering advice and examples of how we can look for opportunities in the least likely places. Watch the 5-minute video to hear Lorsch’s advice to the graduates—and all future scientists—to venture into the unknown in search of the next big advance in biomedical research. Cellular mesenchymal epithelial transition (C-MET) gene copy number variation in gastric adenocarcinoma: A pilot search for new marker for targeted therapy in HER-2/neu resistance.
Abstract Increasing HER-2/neu resistance in gastric carcinoma has encouraged search for new biomarkers for targeted therapy. Cellular mesenchymal epithelial transition (C-MET) is one such tyrosine kinase inhibitor proposed for personalized salvage treatment. We determined frequency of C-MET gene copy number variation (CNV) by Fluorescent in-situ hybridization (FISH) in gastric adenocarcinoma (GAC) and sought its correlation with conventional clinicopathologic parameters. Dual-coloured FISH was done on 32 GAC cases. C-MET gene and centromere 7 signals were counted under fluorescent microscope and ratio was calculat...

In search of a comprehensible set of endpoints for the routine monitoring of neurotoxicity in vertebrates: sensory perception and nerve transmission in zebrafish (Danio rerio) embryos.
Abstract In order to develop a test battery based on a variety of neurological systems in fish, three sensory systems (vision, olfaction, and lateral line) as well as nerve transmission (acetylcholine esterase) were analyzed in zebrafish (Danio rerio) embryos with respect to their suitability as a model for the screening of neurotoxic trace substances in aquatic ecosystems. As a selection of known or putative neurotoxic compounds, amidotrizoic acid, caffeine, cypermethrin, dichlorvos, 2,4-dinitrotoluene, 2,4-dichlorophenol, 4-nonylphenol, perfluorooctanoic acid, and perfluorooctane sulfonic acid were tested in the...

De novo transcriptome analysis to search for sex-differentiation genes in the Siberian sturgeon.
Abstract The sturgeon family includes many species that are lucrative for commercial caviar production, some of which face critical conservation problems. The purpose of this study was to identify genes involved in gonadal sex differentiation in sturgeons, contributing to our understanding of the biological cycle of this valuable species. A high-quality de novo Siberian sturgeon gonadal transcriptome was built for this study using gonadal samples from undifferentiated fish at 3, 5, and 6 months of age; recently sex-differentiated fish at 9 months of age; and immature males and females at 14-17 months of age. Undif...

How to Stay Focused On the Search for Inner Freedom
The terms “being present” and “living in the now” have become clichés on the spiritual path and in the nondual tradition that has become increasingly popular in recent years. We speak of the power of now, the timeless moment, and that “there is only now.” In the positive thinking movement, we released sin as a stumbling block, then replaced it with the charge of negative thinking. Now our greatest put-down is the accusation of being distracted and therefore not present to what is. I am being slightly facetious, but the essential point remains: The key to awakening is in the awareness of what is. This is true ...

Nutrition and neurodevelopment: the search for candidate nutrients in the Seychelles Child Development Nutrition Study
Neurotoxicology. 2020 Dec;81:300-306. doi: 10.1016/j.neuro.2020.09.021. Epub 2020 Oct 14.ABSTRACTThis review examines the role of nutrients in child development and outlines the key nutrients identified as potentially important to neurodevelopment among high fish consumers in the Seychelles Child Development Nutrition Study (SCDNS). It describes the clinical assessment of these nutrients in the blood and breast milk samples collected from the cohort of 300 pregnant women who were recruited, at their first antenatal visit, on the SCDNS. These key nutrients include the long chain polyunsaturated fatty acids (LCPUFA), docosoh...
